Formulation and In-Vitro Evaluation of Bio Adhesive Buccal Tablet of Venlafaxin HCl

A B S T R A C T
Venlafaxine is an antidepressant which belongs to the class of Serotonin-nor epinephrine reuptake inhibitor (SNRI). In the present study hydrophilic bioadhesive buccal tablets of venlafaxine were formulated by using different polymers like Carbopol 934 and carbopol 940 and found that these can be successfully employed as a bioadhesive controlled released polymers. FTIR Studies performed for the drug and excipient, proved that there is no incompatibility. The powder blend of the formulations was evaluated for precompression parameters and it was found that the flow properties of the blend are good. The post compression parameters for all the formulations also found to be within limits. Slow, controlled and complete release of Venlafaxine HCl over a period of 9 hours was obtained from matrix tablets formulated employing carbopol 940 (F5 Formulation) with 93.45 % drug release and the mechanism of drug release is by non-Fickian diffusion. Among all the formulation F5 was found to be best and thus optimised. This optimized formulation is further subjected to evaluations like ex-vivo residence time, surface pH, bioadhesive strength and swelling index, and the results of these tests gave promising results as a bioadhesive drug delivery system.
