Determining the Best Poly Ethylene Glycol as Solid Dispersion Carrier for Improvement of Solubility and Dissolution Rate of Ofloxacin

A B S T R A C T
The present investigation was to find out the best polyethylene glycol (PEG) as carrier to increase solubility of poorly soluble drug by taking Ofloxacin as model drug. PEG 1500, 3350, 4000, 6000, 8000 and 20000 were studies as carrier for solid dispersions. The phase solubility behavior of Ofloxacin in different concentrations of PEG 1500, 3350, 4000, 6000, 8000 and 20000 in distilled water was obtained at 37°C. The solubility of Ofloxacin was found to enhance with increased amount of PEG 1500, 3350, 4000, 6000, 8000 and 20000.  The Ofloxacin solid dispersions with PEG 1500, 3350, 4000, 6000, 8000 and 20000 were prepared using 1:1 and 1:2 (Ofloxacin:PEG) ratio by hot melt method. The compatibility of Ofloxacin with PEG was checked by differential scanning calorimetry (DSC) and Fourier transform infrared (FTIR) spectroscopy. The FTIR spectroscopic studies revealed that there is no chemical interaction and Ofloxacin was found stable. The solid dispersions prepared with PEG 6000 exhibited enhanced dissolution rate of Ofloxacin compared with other PEGs.
