Analytical Method Development and Validation by RP-HPLC for Simultaneous Estimation of Lansoprazole and Domperidone in Combined Tablet Dosage Form

A simple, Accurate, precise method was developed for the simultaneous estimation of the Lansoprazole and Domperidone in Tablet dosage form. Chromatogram was run through Inertsil ODS C185 µm (4.6 mm 250 mm). Mobile phase containing Phosphate buffer and Acetonitrile in the ratio of 55B:45A was pumped through column at a flow rate of 1 ml/min. Buffer used in this method was 0.01N KH2PO4 pH 4.6 buffer. Temperature was maintained at ambient. Optimized wavelength for Lansoprazole and Domperidone was 274 nm. Retention time of Lansoprazole and Domperidone were found to be 3.9 min and 2.3 min. % RSD of the Lansoprazole and Domperidone were found to be 0.18 and 0.39 respectively. %Recover was Obtained as 98.0% and 102.0% for Lansoprazole and Domperidone respectively. LOD, LOQ values are obtained from regression equations of Lansoprazole and Domperidone were 2.95 and 3.09 and 9.87 and 10 respectively. Regression equation of Lansoprazole is y = 18589.x + 37248, and of Domperidone is y = 1581.x + 31392.
