A simple precise and accurate reverse phase high-performance liquid chromatography (RP-HPLC) method has been developed for the simultaneous estimation of Darunavir and Ritonavir. Chromatographic separation was achieved Inertsil ODS C18 column (4.6 x 100mm, 5mm) using the mobile phase consisting of Phosphate buffer 3.5 pH and  Acetonitrile and Methanol in the ratio of 30:70. The mobile phase was pumped at a flow rate of 1.0 mL/min and detection was done by UV detector at 220 nm. The retention times were 2.785 min for Darunavir and 2.105 min for Ritonavir. The linearity ranges for Darunavir and Ritonavir were 120-600 µg/ml and 20-100 µg/ml respectively with correlation coefficient 0.999. The proposed method was found to be simple, accurate, precise and reproducible and could be applied for routine quality control analysis for simultaneous estimation of Darunavir and Ritonavir in pharmaceutical dosage forms.
